FASTA

The Insulin receptor is made up of two chains, meaning it has two subunits and alpha and a beta. These can be subdivided into long and short isoforms.

The receptor has insulin activity by the ability to bind insulin and has tyrosine kinase activity.

It is a tetramer of 2 alpha and 2 beta chains linked by disulphide bonds. The alpha chains contribute to the formation of the ligand-binding domain, while the beta chains carry the kinase domain. Interacts with SORBS1 but dissociates from it following insulin stimulation.

The short isoform has a high affinity for insulin; it also mediates the metabolic functions of insulin. It can activate PI3K by binding to the p85 regulatory subunit. When present in a hybrid receptor with IGF1R it can bind IGF1.

The long isoform, are activated with a high affinity by IGF1, with low affinity by IGF2 and not significantly activated by insulin in comparison to the short isoform.

The short isoform is believed to be a much better form to bind insulin in.

Isoform long and isoform short are expressed in the peripheral nerve, kidney, liver, striated muscle, fibroblasts and skin tissue.
